I've experienced the EK/QF thing before, you have to be on a full EK itinerary with both flights EK numbers (Not even one EK number and one QF number) to get EK upgrade done either online or at the lounge prior.
You can pay cash on board but last time I enquired SIN-MEL it was crazy, i.e. AUD 1,800 crazy.
Sorry to be bearer of bad news but it will not work. Had same question last year, was booked in EK Y on a QF flight number. As usual one cannot pay or bid for any upgrade online. I attempted to do so at check-in. EK staff at CDG attempted to process cash upgrade at check-in desk but 'computer said no' as it was not an EK flight number, apparently. I scoured these forums and elsewhere and have not found any evidence that it can be done.

Hi all - I'm booked at the very back of the bus on EK shortly, all on a QF flight number (EK407 + connections to Europe, return).
The EK website says "cannot purchase an upgrade" but has anyone had any luck either calling them or asking at the airport?
Would being QF platinum make any difference to them?

Try asking at the airport. The usual rules don't apply to airport upgrades.
