I got one last time I flew SQ in J, a year or two ago; not only do they certainly do ozzy ExpressPath cards, they're colourful SQ-branded paper ones that look a bit different to normal ones. That's why I remember it with certainty!
In any case, if it's changed (unlikely!), I've always found the staff member at the departure queue checkpoint to be accomodating if I didn't have an ExpressPath card, but showed them a J ticket on a premium carrier. You'll definitely be fine.
Note ExpressPath now only gets you fast-tracked through security; immigration at MEL is one big zoo, funnelled through the automated ePassport gates.
Jay - worth noting that there is an express path lane that takes you to the front of the of ePassport gates queue. There's a sign and a left turn as soon as you enter the roped area of the immigration hall.
