FLying Perth/Syd on Qantas then to Vancouver on Air Canada all in Business. All booked on one ticket. WIll I be able to get the bags checked through to Vancouver?

Qantas and Air Canada have an interline agreement, so yes your bags can be tagged through to YVR.

Yes, you can. I did this in December checking in from Qantas in Melbourne and connecting in Sydney to AC. It was very easy!
On the return leg, we were given QF boarding passes in YVR. Easy!

I assume this inter-airline agreement does not relate to lounge access at syd, as QF is OW and AC is *A?

Interline arrangements are generally referring to things like selling seats on planes and checking of bags etc.
Things like lounges (outside of alliances) are done airline to airline. If AC wanted passengers to have access to the QF lounge they would come to an agreement... but they don't really need to, as they have 2 star alliances lounges at Syd. If you look Perth for example, many non OW airlines use the QF lounge.

Most likely, you will get access to the KoruClub at SYDi or the KrisFlyer at SYDi.
Both of these are better than QFi SYD J lounge. And both are *A member lounges.
Though if you are QF WP/P1, then you would not get access to the F lounge, as you will need a QF/EK F BP.
I have done the old KoruClub, ugh, but that ones closed now, the new KoruClub should be great.
Have done the KrisFlyer in SYD, that ones not bad, at least you get views to the outside quite well.
QFi SYD J lounge has no proper view to the outside, your view is blocked by the airconditioning ducts. But you would not get access here either. And yes, I have visited this one too. If you were flying QF J and have nothing else, the QFi SYD J lounge is ok.
If you have other choices, go to the other choice, rather than the QFi SYD J lounge. Apart from the QFi SYD F lounge this time round, of course.

Noting that QFWP/P1 also have access to F lounge when flying OW carriers.
In the relation to the OP, QF status is irrelevant on the second leg as he is flying AC.
As he is flying AC business, he will have access to either/or/both star alliance lounge at the airport. The AC page has both listed, but not QF.

PS, you do get access to the new QFd PER J lounge though, as your originating flight from PER will be a domestic one.
QFd PER is the most newly refurbished domestic lounge QF has.
Get there as early as you can!
Go to gate 15 at QFd SYD terminal on arrivla to SYD... don't go out into the open area first. Ask if you can get the QF bus, if yes, you are lucky, if not, you can get across to QFi SYD terminal by the 400 bus, or train, or taxi.

I used the Qantas Business lounge in Melbourne, as my first leg was a QF flight in J, so as above go straight to the lovely new Business lounge in Perth. When we got to Sydney, we did the transfer at gate 15, no questions asked. As our boarding passes were printed on Qantas paper maybe thats what snuck us in, so hopefully you can transfer there as its super easy.
In the International airport at SYD, we used the new AirNZ lounge which is great. Once you cross the pacific though its all downhill from there. In fact, the AC lounge in Vancouver is about on par with the Qantas Club in Launceston- dull.
