You can check the loads through ExpertFlyer (sorry I don't have a subscription, although you can get a 4-day trial). I reckon QF would have already chalked-up plans to put 380 on JNB if it was sustainable.

Can't see it ever getting an A380 - Qantas doesn't have enough and I suspect not enough demand.

Qantas 787-9 will be the perfect option to replace 747-400 for SYD-JNB/SCL route, or hand SCL over to LATAM (they're going to use 787-9 anyway)

I flew this route one way every three to four weeks for work, the flights were often busy but mainly full during school holidays, with services 6 days a week. A daily A380 would probably be overkill, I believe, so a reduction in the number of weekly services may have to occur for it to be viable..

The Qantas graphic for Project Sunrise (ultra-ultra long range aircraft shopping), shows SYD-JNB as one of the target routes. Well, at least the suggested timing fits with the last of the 747ERs going.
